Before SDS- PAGE, DTT can effectively restore the protein disulfide bond in sample buffer. DTT can also be used to restore the cross-linking agent N,N '-double (propylenoyl) - disulfide bond bridge to decompose the polyacrylamide gel matrix. Both the irritant and toxicity of DTT are lower than 2-mercaptoethanol. Usually, the concentration required for DTT (100mM) is one in seven of the concentration of 2-mercaptoethanol (5% v/v, 700mM).
