Features and Application of Guanidine Thiocyanate
Features
Guanidine Thiocyanate is a chemical compound used as a general protein denaturant, being a chaotropic agent, although it is most commonly used as a nucleic acid protector in the extraction of DNA and RNA from cells.
Applications
Guanidine Hydrochloride is useful for the denaturation and subsequent refolding of proteins. This strong denaturant can solubilize insoluble or denatured proteins such as inclusion bodies and be used for the recovery of periplasmic proteins. This can be used as the first step in refolding proteins or enzymes into their active form. Urea and dithiothreitol (DTT) may also be necessary.
It is also used in the isolation of RNA to dissociate the nucleoprotein into its nucleic acid and protein moieties.
It is used for the lysis of colorectal adenocarcinoma cell line prior to RNA isolation.
It is used as a PCR inhibitor in triplex mitochondrial DNA (mtDNA) qPCR assay.
It is used for DNA extraction from cerebrospinal fluid (CSF).
