The fluorescein commonly used in immunofluorescence histochemical staining are FITC, TRITC,RB200,Texas red,PE,Cyanine(Cy3、 Cy5).In addition, there are some new fluorescent dyes, such as quantum dots.
FITC is stable, soluble in water and ethanol, and can bind to proteins. It is the most commonly used fluorescent probe to detect proteins in tissue and cells. It can also be labeled with antibodies, which can be used for single or multiple staining of F immunohistochemistry. The disadvantage is that it is easy to quench under light, and easy to be affected by white hair fluorescence. The maximum excitation wavelength is 490m and the maximum emission wavelength is 525nm, showing yellow green fluorescence.
