BCA is a widely used protein quantitative method recently. Its principle is similar to Lowery method in protein quantification, that is, in alkaline environment, the protein and Cu2 + complex combine to reduce Cu2 + to Cu1 +. BCA combines with Cu1 + to form a stable purple blue complex, which has a high light absorption value at 562nm and is directly proportional to the protein concentration. Compared with Lowery's method, BCA protein determination method has high sensitivity, simple operation, good stability of reagents and their formed color complexes, and little influence by interfering substances. Compared with Bradford method, BCA method has the advantage of no detergent.
